In vitro study | When N2AB-1 and glioma cells were exposed to various concentrations of MPTP, their morphology did not change, and at the same time, the proliferation of C6 glial cells was not affected. MPTP promotes apoptosis and Tau phosphorylation in human neuroblastoma M17 cells, and MPTP significantly increases the phosphorylation level of Tau protein at ser262. MPTP causes elevation of intracellular α-synuclein levels in human neuroblastoma M17 cells. MPTP promotes the phosphorylation of Tau in the brain by activating PKA and GSK3β. |
In vivo study | The number of tyrosine hydroxylase-positive neurons was reduced in the substantia nigra pars compacta of MPTP-treated mice. In the mouse midbrain, MPTP reduces the expression and activity of thioredoxin type 1 reductase and reduces thioredoxin type 1 reductase-positive cells in the substantia nigra pars compacta. MPTP can cause neurochemical, behavioral, and histopathological changes in humans and non-human primates, similar to the symptoms of Parkinson's. Compared to primates, rodents are less sensitive to MPTP. MPTP can be administered through a variety of routes, such as feeding method, positioning injection method, but the most common, repeatable route is systemic administration, including subcutaneous, intravenous, intraperitoneal, intramuscular injection. MPTP is a lipophilic protoxin that permeates the blood-brain barrier. It must enter the brain and MPTP is converted to 1-methyll-4-phenylpyriridine by monoamine oxidase B. MPTP has been shown to be toxic to dopaminergic neurons in the nigrostriatal system of humans, monkeys and mice, continuously depleting dopamine and producing its metabolites in the striatum. |
